To quantify the area of the growth plate composed of cartilage (which stains red after Safranin O/Fast Green staining), images were imported into Adobe PhotoShop. The area of the Safranin O stained cartilage growth plates was measured in a double-blinded manner by two independent investigators. At post-natal day 8 (P8) the midpalatal suture complex is made up of three elements: the bony palatine processes of the maxillae, the cartilage growth plates that cap the ends of the palatine processes (red arrows), and the fibrous interzone (asterisk) that separates the growth plates (Fig. 1A).
